Abstract

The importance of high quality of seed is aimed to improve the production of potato. Nowdays, the potato farmer generally doesnt use high quality of potato seed because the price is relatively expensive and it quite hard to find. This research aimed to get sterile explant and regenerate it in in Vitro technique. This research used 5 levels of coconut milk, they are 100, 150, 200, 250, and 300 ml of coconut milks. The result of the rsearch showed siginificantly different respons to the numbers of parameters they are, early shoot formation, numbers of shoot, and shoot length. Meanwhile the paramaters that didn't show non-significantly different were early root formation, leaves length, numbers of Nodes, and numbers of leaves. The media that contained of higher concentration of cytokinin than auxin stimulates shoot formation, in contrary, the high concentration of auxin stimulates root formation. The cytokinin level in coconut milk was able to stimulate shoot formation nicely. Based on the six treatments that were made, the best respon was showed by the treatment of s5 (ms0 + 3 ppm(s) of BAP + 0.01 ppm(s) of NAA + 250 ml of coconut milk per liter of media.
