Abstract
Iles-iles (Amorphophallus muelleri Blume) is a native Indonesian tuber, originating from the Araceae tribe containing mannose and mannan called glucomannan with levels between 40-65% which can be used as a source of carbohydrates other than rice. Conventionally, in the multiplication of iles-iles plants, there are many obstacles that need to improve genetic diversity by tissue culture. The purpose of this study was to determine the type of explants and the concentration of coconut water which was right in inducing iles-iles shoots in vitro. This research was conducted in December 2018 until May 2019 at the Jember State Polytechnic Tissue Culture Laboratory. The experimental design was arranged in factorial randomized block design (RBD), each treatment repeated 3 times. The first factor is the type of explants which consists of the use of types of explants including leaves, petiole, callus. The second factor is the addition of coconut water to a culture media with a concentration of 150; 250; 400 ml/L. The results showed that leaf stalk and callus types were the best explants in inducing shoots of iles-iles. Concentration of coconut water 150 ml/L gives optimal results for shoot height, 400 ml/L concentration of coconut water is less optimal for all parameters. While the concentration of 250 ml/L provides optimal results for all observed parameters.
