Abstract
The production of potatos in Indonesia are decrease every year. It caused by limited supply of qualified seeds. The supply of qualified seeds can increase by tissue culture technique with induction of micro tuber. This research aims to determine the formation of potato micro tuber on in vitro environment by optimizing the concentration of nitrogen (NH4NO3) and auxin type. This research was conducted at Tissue Culture Laboratory of State Polytechnic of Jember in July 2018 to January 2019. It was applied Completely Randomized Design with 2 factors. The first factor was concentration of nitrogen consisted 3 levels, there were 10 ml/l (N1), 20 ml/ l(N2), and 30 ml/l (N3). The second factor was auxin type 0,1 ppm IAA (A1), 0,1 ppm NAA (A2) and 0,1 ppm IBA (A3). The data was analyzed by f Test (ANOVA) and followed by Duncan’s Multiple Range Test (DMRT) 5%. The result of this research shows that interaction between concentration of nitrogen and auxin type is significant on early tuber , number of tuber, size of tuber and gross weight of tuber. The best combination treatment on parameter diameter of tuber and gross weight of tuber is 20ml/ NH4NO3 with auxin 0,100m NAA, on parameter early tuber is 10 ml/l NH4NO3 with 0,1 ppm IAA, and on parameter number of micro tuber is 30 ml/l NH4NO3 with 0,1 ppm IBA.
