Abstract

This research aims to determine the effect of auxin type and concentration of BAP on Induction of Callus of Iles-Iles (Amorphophallus muelleri Blume.). This research was conducted at Tissue Culture Laboratory of State Polytechnic of Jember in September 2018 to January 2019. This research was using Randomized Complete Block Design (RCBD) wtith 2 factors. The first factor was auxin type consisted 2,4 D 1 mg/l; 2,4 D 2 mg/l; NAA 1 mg/l; NAA 2 mg/l; IAA 1 mg/l; and IAA 2 mg/l. The second factor was concentration of cytokinin consisted BAP 0 mg/l; and 1 mg/l. The data was analyzed by ANOVA and followed by DMRT 5%. The result of this research shows that interaction between auxin type and concentration of BAP is significant on callus biomass, root formation rate and number of roots. The best treatment is NAA 2 mg/L + 0 mg/L BAP and significant effect on callus biomass of 1,14 gram, root formation rate of 42 days after innoculation and number of root of 5,68. The interaction between auxin type and concentration of BAP is non significant effect on callus early formation, bud early formation, percentage of callus formation, color of callus, texture of callus, number of bud and height of bud. NAA is the best auxin on callus biomass, root formation rate and number of root. No BAP addition is the best treatment on callus biomass, root formation rate and number of root.
