Abstract

Black potato (Plectranthus rotundifolius) is an alternative food source that can support food diversification activities. The existence of black potatoes is still rare, so it needs to be preserved in order to become a source of biodiversity. Therefore it is necessary to propagate plants in vitro, to maintain the presence of black potatoes. This research was conducted by adding Kinetin and IBA to stimulate the multiplication of black potato buds (Plectranthus rotundifolius). This research was conducted in September 2019 to February 2020 at the Green House and Tissue Culture Laboratory, Jember State Polytechnic. The experimental design used a completely randomized design (CRD) with 2 factors and 5 replications. The first factor is the addition of Kinetin 2 ppm (K1), 4 ppm (K2) and 6 ppm (K3). The second factor is the addition of IBA 0.5 ppm (B1), 1 ppm (B2) and 1.5 ppm (B3). The results showed that the addition of kinetin with a concentration of 2 ppm (K1) can stimulating the appearance of shoots and the addition of kinetin with concentration of kinetin 6 ppm (K3) gave the best results for the diameter of the stem. IBA concentration of 0.5 ppm (B1) is optimal for stimulating the number of segments, root length and shoot height. The combination of kinetin 2 ppm + IBA 0.5 ppm (K1B1) is effective to stimulate the number of shoots, number of roots and shoot height. The combination of kinetin 4 ppm + IBA 1.5 ppm (K2B3) is optimal for stimulating the appearance of roots.
