Abstract

Sorghum plays a role as an alternative food crop that is still in the introduction and development stage in Indonesia. The in vitro culture approach is one effective technique for developing sorghum plants in a non-conventional manner and producing more than one shoot. The success of multiplication is supported by the administration of growth regulators. This study aims to analyze the effect of adding the growth regulators BAP (Benzil Amino Purin) and NAA (Naphthalene Acetic Acid) and a combination of both on the multiplication of sorghum (Sorghum bicolor L.) shoots in vitro. This study was conducted from June to October 2025 at the Tissue Culture Laboratory, Jember State Polytechnic. This study used a completely randomized factorial design (RALF) with 9 treatment combinations and 3 replicates. The first factor was the concentration of BAP, consisting of 1 ppm, 2 ppm, and 3 ppm, while the second factor was the concentration of NAA, consisting of 0 ppm, 0.5 ppm, and 1 ppm. The observation variables included sprouting time, number of sprouts, sprout height, number of roots, root length, and number of leaves. The results showed that there was an interaction between the treatment combination of 1 ppm BAP and 0 ppm NAA on the time of sprouting (3.00 days). The treatment of 1 ppm BAP had a very significant effect on sprout height (14.3 cm). Meanwhile, the treatment of 0 ppm NAA had a very significant effect on root length (2.40 cm).
