Abstract

This study aims to find a combination of MS medium concentration and coconut water effective to optimize the growth of Prancak 95 varieties of tobacco shoots. This research was conducted at the plant tissue culture laboratory, State Polytechnic of Jember from September 2018 to January 2019. This study was designed with a factorial complete randomized design. The first factor is the concentration of MS medium which consists of two levels i.e. 1 MS and ½ MS. The second factor is the concentration of coconut water which consists of four levels i.e. 5%; 10%; 15%; and 20%. The results showed that the combination of MS medium concentration and coconut water had an effect on the average root growth rate of 9.4 HST, number of roots 13.4, root length 82 mm, number of leaves 12.4 and shoot height 90 mm. Single factor The best MS medium concentration is 1 MS (full MS) while the concentration of coconut water 10% and 15% is the best concentration for the growth of prancak 95 varieties of tobacco shoots.
