Abstract

Orchid is an ornamental plant commodity that plays an important role in both domestic and foreign markets. The orchid commodity is in fourth place with a total of around 2.66% of the total national cut flower production. The success of in vitro plant propagation in the laboratory really depends on the final stage of propagation, namely acclimatization. Acclimatization is the stage of adapting plantlets from the in vitro environment to the environment outside the bottle. Acclimatization is obtained by the growing medium. This study aims to determine the effect of planting media on the addition of Dendrobium sp. on the acclimatization process. This research was conducted on 19 October to 18 December 2017 at the Tissue Culture Laboratory and Green House of Jember State Polytechnic. The experiment was prepared using a non-factorial completely randomized design (CRD) with acclimatization growing media treatment consisting of 3 treatments using fern root media (M1), wood charcoal media (M2), a mixture of fern root media + wood charcoal media (M3), repeated 6 times. The decomposition variables of: plant height, number of leaves, and leaf area, all data were analyzed using ANOVA, if there is a significant difference, then continue with the DMRT level of 5%. The results showed that the type of planting medium had no significant effect on all the observed variables. Key word: Dendrobium orchid, aclimatization, a type of media planting
