Abstract

This research aimed to determine the effect of BAP (6-Benzyl Amino Purine) and NAA (Naphthalene Acetic Acid) on the growth and number of shoots in vanilla (Vanilla planifolia Andrews.) through in vitro culture. The research was conducted from February to June 2025 at the Plant Tissue Culture Laboratory, Politeknik Negeri Jember, using a factorial completely randomized design (CRD) with two factors: BAP (0, 1, and 2 mg/L) and NAA (0, 0.5, and 1 mg/L), resulting in nine treatment combinations with three replications. The observed parameters included the time of shoot emergence, number of shoots, and shoot height. The results of the study showed that the application of BAP had no significant effect on all observed parameters. The application of NAA had a significant effect on the number of shoots but no significant effect on the time of shoot emergence or shoot height, with the best results obtained from the treatment without NAA (N0). The combination of BAP and NAA had a significant effect on the number of shoots but no significant effect on the time of shoot emergence or shoot height. The best combination was found in B1N0 (BAP 1 mg/L + NAA 0 mg/L), which produced six shoots
