Abstract

Black potato is an alternative food containing high carbohydrates (18.92%), drought tolerance, and wide adaptability. However, the low availability of quality seeds is a major problem. Therefore, the preparation of quality seeds on a large scale through tissue culture must be carried out. Through this technique, seeds (tubers and micro cuttings) were pathogens-free, similar, and independent of season. However, there is a critical phase in tissue culture, namely acclimatization. The growing media during acclimatization has an important role to support plantlet growth. This study aimed to determine media type and nutrients on the growth of black potato during acclimatization. The experiment was conducted from April to August 2021 at Jember State Polytechnic. The experiment was arranged in a factorial completely randomized design with 2 factors and 3 replications. The first factor was planting media, namely cocopeat+compost and rice husk ash+compost, while the second factor was the nutrient concentration of AB Mix which consisted of 7 ml/l, 11 ml/l, 15 ml/l, and 19 ml/l. The results showed that rice husk ash+ compost with AB Mix 7 ml/l nutrients gave the best effect on the leaves area (4.2 cm2). It has a high content of silica, as well as phosphorus and potassium which can increase the photosynthesis rate. The growth characteristics of black potato (plant height, number of leaves, growth rate) tended to be better in the media with rice husk ash added. Meanwhile, the application of AB Mix 7 ml/l had a better tendency toward the growth of black potato.
